A feasable route will be to go directly to Gangtok from Kolkata via NH 34. From here you can go to Darjeeling (approx 100 km). From Darjeeling you can go to Siliguri.

From here, if you are planning a road trip, you'll have to go to a place called Panitanki (Indian side of the border), pass through customs which is open 24X7 and takes around 10-15 minutes. Once you clear the customs, there's a 1 km walk that will lead you to Kakarbhitta, which is the Nepalese side of the border. Clearing the customs here will again take some 10-15 minutes. There are a number of tourist buses and local buses that will take you to Kathmandu. Going for the luxury tourist buses is highly recommended as the roads are in bad conditions and travelling in local buses might be torturous. The fare in the luxury buses should be around NPR 1200-1500.

If you wish to fly to Kathmandu, then you'd have to come to Bagdogra Airport from Siliguri.
